چکیده

INTRODUCTION Mechanical impact loading is known to cause cell death and to lead to the onset and progression of osteoarthritis [1]. In order to understand why in-situ cells die readily following impact loading but remain unaffected when the same load is applied at a slow rate, we used a Finite Element model of articular cartilage and chondrocytes to study the in-situ cell mechanics, which cannot be measured experimentally, especially at impact loading rates, with present day technology. The objective of this study was to investigate the effects of strain rate on in-situ chondrocyte mechanics. We hypothesized that tensile strain rates on cell membranes hold the key to explaining cell deaths at high loading rates.

Measuring microscale strain fields in articular cartilage during rapid impact reveals thresholds for chondrocyte death and a protective role for the superficial layer.

Articular cartilage is a heterogeneous soft tissue that dissipates and distributes loads in mammalian joints. Though robust, cartilage is susceptible to damage from loading at high rates or magnitudes. Such injurious loads have been implicated in degenerative changes, including chronic osteoarthritis (OA), which remains a leading cause of disability in developed nations. Despite decades of rese...

Mechanical Behavior of in-situ Chondrocyte at Different Loading Rates: A Finite Element Study

Previous findings indicated that in-situ chondrocytes die readily following impact loading, but remain essentially unaffected when loaded at the same magnitude but with a slow (non-impact) loading rate. The current study was aimed at identifying the causes for cell death in impact loading by quantifying chondrocyte mechanics when cartilage was compressed nominally by 5% at different loading rat...

Mechanical loading of in situ chondrocytes in lapine retropatellar cartilage after anterior cruciate ligament transection.

The aims of this study were (i) to quantify chondrocyte mechanics in fully intact articular cartilage attached to its native bone and (ii) to compare the chondrocyte mechanics for cells in healthy and early osteoarthritis (OA) tissue. We hypothesized that cells in the healthy tissue would deform less for given articular surface pressures than cells in the early OA tissue because of a loss of ma...

A Review of the Combination of Experimental Measurements and Fibril-Reinforced Modeling for Investigation of Articular Cartilage and Chondrocyte Response to Loading

The function of articular cartilage depends on its structure and composition, sensitively impaired in disease (e.g. osteoarthritis, OA). Responses of chondrocytes to tissue loading are modulated by the structure. Altered cell responses as an effect of OA may regulate cartilage mechanotransduction and cell biosynthesis. To be able to evaluate cell responses and factors affecting the onset and pr...

Oscillating fluid flow regulates cytosolic calcium concentration in bovine articular chondrocytes.

Mechanical loading is a well-known regulator of cartilage metabolism. This suggests that a loading-induced physical signal regulates chondrocyte behavior. Previous studies have focused on the effects of steady fluid flow on chondrocytes. In contrast to steady flow, loading induced fluid flow occurs in an oscillatory pattern and includes a reversal of flow direction with each loading event. In t...
